Omar Sy partage son succès avec Hélène, une femme qu’il a connue alors qu’il était encore un comédien méconnu du grand public en France. Ils se sont fréquentés depuis 1998, mais ne se sont mariés qu’en 2007. Ils sont parents de cinq enfants dont la plus jeune est née en octobre 2017.
